The aim of the article is to determine the importance of an integrative approach to identify and analyze the phases or stages of the example of chronic sclerotic disease (CSD). The proposed concept of CSD is to use the diagnostic methods before sclerotic period, based on an analysis of indicators of intensity of the functional systems responsible for collagen formation and collagen lysis taking into account the potential of genetically caused features of sclerosis. Thus, special attention is paid to pre-disease stage including the history and molecular genetic studies which predict the nature of the possible diseases and determine the risk of formation of CSD. The proposed methodology and techniques, as well as obtained with their help data define the possibility of risk groups identification, the direction and volume of the survey.

The relationship between the degree of hydroxylation and oxidative deamination of e-NH2-groups of lysine and hydroxylysine in collagen, and the thermal stability of this protein’s sub-molecular structures in the skin of Wistar rats in their postnatal ontogeny was in vitro studied. It has been shown that, rising towards the age of 3 months, the content of free e-NH2-groups in collagen continuously remains constant, while that of free aldehyde groups (COH-groups), starting from 1-month steadily decreases with age. Accordingly, intermolecular cross-linking in collagen’s sub-molecular structures is reduced during the period between 1 to 3 months, and afterwards continuously increases up to 24 months of age. The content of hydroxyproline in collagen is continuously reduced in postnatal ontogenesis. The combined effect of both effects leads to a decrease in the thermal stability of collagen sub-molecular structures in the skin during the period from 1 to 3 months, and subsequently rises up to 24 months old.

Age-related changes in concentrations of the major connective tissue macromolecules – collagen, elastin, separate fractions of glycosaminoglycans – hyaluronic acid, chondroitin sulphates, heparan sulphate, and dermatan sulphate, as well as their proportion in both the skin and aorta were studied. An age-related increase of collagen content in both the skin and aorta and at the same time a decrease of elastin content were shown in these tissues. The content of hyaluronic acid, chondroitin sulphates, heparan sulphate and dermatan sulphate increases in the skin with age. Contrarily, in the aorta, a decrease in concentrations of hyaluronic acid and dermatan sulphate takes place with age, while those of chondroitin sulphates and heparan sulphate increase.
